Protein extraction from rat tissue using Omni Bead Ruptor 4

The Omni Bead Ruptor 4 (BR 4) is a compact bead mill homogenizer designed to efficiently lyse tissues for extraction of intracellular compounds, including proteins for downstream molecular analysis. In this study, protein was extracted from various rat tissues, including muscle, brain, liver, and heart using total mass inputs ranging from 22 – 30 mg; tissues were processed using different instrument parameters for robust sample homogenization, as observed by visual inspection of completion. Protein concentrations were quantified, normalized, and assessed using nanodrop and by SDS-PAGE. For labs looking for a lower throughput, but multi-sample bead mill homogenizer, this unit was more than proficient at extracting proteins from up to 4 samples-at-a-time from various tissue types effectively. With a short processing time, robust yields were obtained with a broad molecular weight range as observed by gel electrophoresis, indicating utility for a wide variety of applications.
