Transcriptome research has become increasingly popular to monitor global changes in gene expression. In particular, transcriptome changes between diseased versus healthy states can provide valuable insight in understanding disease mechanisms. The isolation of high-integrity mRNA from tough tissues, such as skin, can represent a significant challenge in research. In this application note, we compare two methods for mRNA extraction from two murine skin types. Our study demonstrates the efficacy of using bead mill technology as a reliable sample preparation method.
Access the full app note for the full protocol, yields, ass well as the electropherogram traces from ear and ventral skin processing on the bead mill, including two different staring input quantity for tough ventral skin, and how they compare to data obtained from older, more manual dounce homogenization (glass mortar & pestle).
