The NEXTFLEX® NGS Cleanup Beads are Solid-phase reversible immobilization (SPRI) beads that efficiently purify nucleic acids using reversible magnetic bead binding. They are ideal for PCR and NGS cleanup, removing unwanted components like soluble contaminants, nucleotides, primers, salts, and adapters. They selectively bind DNA by size with high recovery and consistent results. These SPRI beads can be used manually or in any automated liquid handler, depending on sample throughput.

The NEXTFLEX NGS Cleanup Beads are a cost-effective solution for these applications:

Performance

Figure 1 shows an example of size selection performed with NEXTFLEX® NGS Cleanup and a popular competitor using different bead-to-sample ratios. Results show a predictable and consistent size selection, comparable to competition.

Figure 1. 625 ng of 50 bp DNA ladder (Promega) were purified with equal amount of AMPure® XP (Beckman) and NEXTFLEX® NGS Cleanup and run on 1% agarose gel for comparison.

Figure 2. shows final yields obtained when using same bead-to-sample ratio of either the NEXTFLEX® NGS Cleanup or a popular competitor to cleanup the libraries generated with our NEXTFLEX 16S V4 2.0 kit. Yields of both solutions are not significantly different.

  • What are NEXTFLEX NGS Cleanup Beads used for?

    NEXTFLEX NGS Cleanup Beads are SPRI-based magnetic beads used to purify DNA fragments during NGS library prep or PCR clean-up. They bind DNA (selectively by size) and remove primers, nucleotides, salts, and adapters. Use them in protocols requiring DNA cleanup or size selection.

  • How do NEXTFLEX NGS Cleanup Beads compare to AMPure XP beads?

    NEXTFLEX beads are designed as a drop-in replacement for Beckman Coulter’s AMPure XP. In head-to-head tests, they deliver comparable DNA recovery and size-selection performance at a much lower price.     Yes. By adjusting the bead-to-sample ratio, NEXTFLEX beads can perform DNA size selection (e.g. enriching or depleting fragments above or below certain sizes). Revvity’s data shows predictable, consistent size cuts similar to AMPure XP or SPRIselect.

  • What is SPRI (Solid-Phase Reversible Immobilization) technology?

    SPRI is a DNA purification method using paramagnetic beads coated with a reversible DNA-binding chemistry. When mixed with DNA and a binding buffer, the beads capture nucleic acids. A magnet pulls the beads (with DNA) aside for washing, then DNA is eluted. SPRI beads enable high recovery and scalable cleanup without centrifugation.

  • What DNA fragment sizes can NEXTFLEX NGS Cleanup Beads recover?

    These beads efficiently recover double-stranded DNA fragments >100 bp, with typical recoveries of 70–90% for fragments above 100 bp. By adjusting the reagent ratio, you can target different size ranges (small fragments or large fragment selection) for sequencing libraries.

  • Are NEXTFLEX NGS Cleanup Beads compatible with automated workflows?

    Yes. The beads are fully compatible with standard liquid-handling platforms (e.g. robotic workstations). They work both manually and on automation, offering the same SPRI cleanup protocol.
