Fibroblast growth factor 23 (FGF23), a 32 kDa secreted protein, is a member of the FGF family. More specifically, it is a part of a subfamily that also includes FGF19 and FGF21. FGF23 is predominantly secreted by osteocytes and osteoblasts. The primary function is to regulate vitamin D metabolism and phosphate homeostasis. Secreted levels of FGF23 have been observed to go awry in certain tumor lines resulting in overproduction of FGF23 and hypophosphatemia. Further, elevated levels of FGF23 have been suggested as a biomarker for chronic kidney disease, where an early sign is altered phosphorus metabolism.

AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
