Proteins play a critical role in cellular function, and understanding their expression across individual cells is essential. TotalSeq™ oligo-conjugated antibodies enable simultaneous profiling of protein markers alongside transcriptomes in single cells using techniques like CITE-Seq. By combining protein and RNA readouts, researchers gain high-resolution insights into heterogeneous cell populations, surpassing RNA clustering methods and improving cell type identification.
However, challenges persist in single-cell RNA sequencing (scRNA-seq), where up to 50% of reads can be abundant transcripts which can hinder the detection of low-abundance transcripts. Enter the DepleteX ® Single Cell RNA Boost Kit, based on a novel CRISPR-Cas9-based depletion technology that selectively removes uninformative sequences prior to sequencing, enhancing cell type resolution while reducing reads per cell. In this study, we explore the compatibility of TotalSeq ™ and DepleteX ® in established 10x Genomics workflows, aiming for a more efficient analysis of the cellular transcriptome.
