Antibodies for immunophenotyping with the Cellaca PLX™ image cytometry system.

Primary samples used by researchers in cell and gene therapy, immunotherapy, cell line development, and immuno-oncology are unique and precious due to their biological nature. The Cellaca™ PLX platform, which includes an image cytometry instrument, reagent kits, and software, is designed to increase the efficiency of sample processing, from cell staining to data readout.

The fluorescent labeled antibodies and stains in the Cellaca PLX kits are optimized for ease of use. The pre-defined assays and templates, together with the ready-to-use reagent kits enable flow-cytometry-like data analysis with no need for titrations, extra incubations, washes, or decontamination. These features permit researchers to rapidly move processed samples to downstream applications while preserving sample integrity.

The portfolio consists of:

Single color antibodies
Single color antibodies and isotype controls have been optimized for labeling PBMCs, PBMCs with red blood-cell contamination, and cultured cell lines.

Single surface marker viability kits
Single surface marker kits contain a fluorescently-conjugated antibody for a specific T-cell receptor, a corresponding isotype control, a dead cell identification dye, and a total cell stain.

Two surface marker total cell kits
These kits contain a combination of two fluorescently-conjugated antibodies for specific T-cell receptors, corresponding isotype controls, and total cell dye for identification of nucleated cells. The kits have been designed to work with samples that may contain RBC contamination with the PBMC sample.

Two surface marker dead cell kits
These kits contain a combination of two fluorescently-conjugated antibodies for specific T-cell receptors, associated isotype controls, and a dead cell identification dye. The kits have been designed to work with cell line samples and PBMC samples that have undergone an RBC lysis step and therefore do not have red blood cells in the samples.

Two surface marker viability kits
These kits contain a combination of two fluorescently-conjugated antibodies for specific T-cell receptors, associated isotype controls, and a dead cell identification dye, and a total cell dye for identification of nucleated cells. The kits can be used with cultured cell lines, PBMCs, and PBMCs with red blood-cell contamination.

Three surface marker total cell kits
These kits contain a combination of three fluorescently-conjugated antibodies for specific T-cell receptors, associated isotype controls, and total cell dye for identification of nucleated cells. The kits have been designed to work with samples that may contain RBC contamination with the PBMC sample.

Three surface marker dead cell kits
These kits contain a combination of three fluorescently-conjugated antibodies for specific T-cell receptors, associated isotype controls, and a dead cell identification dye. The kits have been designed to work with cell line samples and PBMC samples that have undergone an RBC lysis step and therefore do not have red blood cells in the samples.

Data from Cellaca PLX System was exported to FCS Express™ Flow Cytometry software for population analysis. Human PBMCs were labeled with anti-CD3 KIRAVIA Blue 520™ and Hoechst. A gate was set on cells that were CD3 and Hoechst positive (shown in A). Further population analysis showed CD3 positive cells that were also CD4-PE and CD8-APC positive (shown in B).
